Biocatalysis breakthrough: new potential in chiral Intermediates

 
Enantioselective Reductive Amination

 

Recombinant and mutagenized amine dehydrogenase has been developed with the help of genetic modification for reductive amination of ketones.

This new enzymevariant catalyses the reductive amination of a variety of carbonyl compounds especially aromatic ketones to enantiopure amines.

A broad library has been created based on the DNA-sequence of the new enzymemutant. A team of molecular biologists and chemists of X-Zyme conducts the screening of this library for the selection of biocatalysts with desired properties.

 

 

 

Properties

 

Important properties concerning the application of the enzyme are the recombination and relatively high stability. Overexpression level: 20% of total cell protein. Activity 17 U/mg. Substrate spectra: aromatic and halogenated aliphatic ketones.

pH-optima: 7.5 (reduction). 

Mg 2+ increases the activity.

1 U corresponds to the amount of enzyme which reduces 1 µmol 4-phenyl-2-butanone to 1-Methyl-3-phenylpropylamine per minute at pH 7.5 and 30 °C
