An antibody specific for WNT3A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyWNT3A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for WNT3A is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of WNT3A bound in the initial step. The color development is stopped and the intensity of the color is measured. Product Overview:The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regµLation of cell fate and patterning during embryogenesis. WNT4 is a member of the WNT gene family, and is the first signaling molecµLe shown to influence the sex-determination cascade. It encodes a protein which shows 98% amino acid identity to the Wnt4 protein of mouse and rat. This gene and a nuclear receptor known to antagonize the testis-determining factor play a concerted role in both the control of female development and the prevention of testes formation. This gene and another two family members, WNT2 and WNT7B, may be associated with abnormal proliferation in breast tissue.
