Test principle:This assay employs a two-site sandwich ELISA to quantitate SMAD3 in samples. An antibody specific for SMAD3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySMAD3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for SMAD3 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SMAD3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

SMAD3 or Mothers against decapentaplegic homolog 3 is a polypeptide that, as its name describes, is a homolog of the Drosophila protein "Mothers against decapentaplegic". It belongs to the SMAD family of proteins, which belong to the TGF? superfamily of modµLators. Like many other TGF? family members, SMAD3 is involved in cell signalling. SMAD3 modµLates signals of activin and TGF?'s.
